You can use variables in the AI Text Generate and AI Transition card instructions, but you cannot use variables in the AI Task Instructions input - variables are only allowed in the "Task Input" part. I like to define my prompts as variables in code cards - it's a larger window and easier to manage long prompts. I can do this for AI Generate and AI Transition cards, but not for AI Tasks.
